Microscope Magnification Recommendations

Please see our recommendations on microscope magnification to accomplish specific tasks, Please refer to the magnification of your particular microscope as this covers microscopes in general. The method to determine your magnification is to multiply the magnification listed on the objective lense (IE, 40X) by the magnification listed on the eyepiece (commonly 10 or 16x) OR the LCD screen (commonly 25x, 25x when purchased from CanineP4.com). 
Depending on your brand of microscope, your results may not be identical to what is displayed below due to build quality and clarity. Magnification without excellent clarity and contrast does not register very good results, and the maximum magnification of your particular microscope may appear blurry as it is as far as it can see.

100X:
Here is a stained sperm slide viewed at 100X total magnification. The sperm cells are not quite visible to the user and appear as many dots. Magnification this low is generally not helpful to the user.

250X
Here the individual sperm cells are more apparent. It is possible to evaluate general motility from a glance at 250X and above, however it is still a birds-eye view. 200-500x can provide a great general overview of the sperm's health visually.

1000X
At 1000X the individual sperm cells are much more apparent. If they are live samples and moving with good velocity, they may dart across the screen fairly quickly. The head and tail are fairly prominent but the mid-piece is hard to identify. 

1500X
1500x yields greater magnification than the prior step, however contrast, and difference in color scale, may be more apparent and require better fine-tuning. Live sperm cells will quickly dart to and fro across your observation area. Make sure your distance is optimum, and light output and filter are set to the best position to view the sperm the best.

2500X
2500x requires a 100x oil objective with oil typically. Results may vary if oil is not used. This high of Zoom may be difficult to tune properly, but once set can give you an excellent image of a complete single cell (depending on density of the sample). Here we can identify the mid-piece and do a much better observation of morphology (study of the form of the cell) to identify things such as detached tails, misshapen heads, and other defects. Sperm cells are best viewed dead or incapacitated at this magnification as it will be hard to see anything clearly while they are moving.
